Diagnosis of Tritrichomonas foetus infection by ELISA and IFA



Abstract
Tritrichomonas foetus(T foetus) is the pathogen causing early embryo death, abortion and pyometra etc in cows. The conventional diagnosis of disease is based on direct smear and culture of vaginal swab from cow. In order to diagnose T foetus, the serological methods of ELBA and IFA were applied to detect the anti T foetus IgG in 40 SPF mice and 21 cows. Twenty seven mice immunized with killed T foetus antigen were alloted to infected-group and 18 mice, control-soup, respectively.
The absorbance of the marine sera of infected-group were 1.4283¡¾0.1574, 1:2901¡¾0.2235 and 1.2727¡¾0.2202 in ELISA at 800- 1,800- and 3,200-fold dilution, respectively. Otherwise, absorbance of the control-group were 0.3627¡¾0.0888, 0.3565¡¾0.0674 and 0.3518¡¾0.0607, respectively. Absorbance of control group at 1,600 dilution of sera was 0.3565¡¾0.0674, and negative in IFA at 18-fold dilution from sera of infected group was 1.1201¡¾0.2817.
Twenty seven mice of infected group were positive in IFA at 2- to 8-fold dilution of sera. Seventeen, 14 and 6 sera from infected mice showed positive reaction in IFA at 18-, 32- and 64-fold dilution, respectively. Twelve, 7 and 2 mice of control group were positive in IFA at 2-, 4- and 8-fold dilution, respectively. No mice showed positive at over 8-fold dilution of sera.
In field application, twenty one bovine sera were collected from local dairy farm and the results of absorbance in ELBA were 0.6100¡¾0.0919, 0.5892¡¾0.0922 and 0.5462¡¾0.1759 at 800-, 1,600 and 3,200-fold dilution, respectively.
This study indicate that serological teat of ELISA or IFA can be used for the diagnosis of T foetus.
